The basis on which the get electrophoresis technique works is the following:
a) Cutting of DNA by restriction endonucleases resulting in the fragmentation of DNA. These fragments are separated by gel electrophoresis.
b) As DNA is negatively charged, it can be separated by forcing it to move towards the anode under an electric field through a medium like agarose.
c) The DNA fragments separate according to their size through sieving effect provided by the agarose gel. Hence, the smaller the fragment size, the farther it moves.
Two ways the products like DNA obtained through this technique can be utilized are: The DNA fragments are used in constructing recombinant DNA by joining them with cloning vectors. DNA is also used for forensic purposes.
